6, Toxicity of melatonin:
Because melatonin is an endogenous substance, it acts through the regulation of the endocrine system and has its own metabolic pathway in the body. 70% ~ 75% of melatonin in blood is metabolized into 6-hydroxymelatonin sulfate form in liver and excreted through urine (80%) and feces (20%).  Another 5%-7% is converted into 6-hydroxymelatonin glucoside acid form, which will not cause the accumulation of metabolites in the body; And its biological half-life is short, in oral 7 ~ 8 h will be reduced to the normal physiological level, so its toxicity is very small. Jack(1967) showed that melatonin soluble dose of 800 mg/kg BW did not cause death in mice. Fu Jianyun et al. (1997) conducted acute toxicity test and mutagenicity test on melatonin in rats and mice, and the results showed that oral LD50 was greater than 10 g/kg BW in rats and mice. Ames test, micronucleus test of bone marrow cells in mice and sperm abnormality test in mice were all negative. Over 3,000 human trials have shown that up to a few grams (thousands of times a healthy dose for maintenance) are taken daily for up to a month with no or little toxicity.
Enzymatic and non-enzymatic reactions in the body may produce free radicals which are closely related to aging. The production and elimination of free radicals in the normal body are in a dynamic balance.
