In vitro studies of nystatin resulted in a significant decrease in buccal epithelial cell adhesion in all six Candida species [1]. Nystatin is an antibiotic that increases plasma membrane permeability to small univalent ions, including chloride ions. Nystatin increases apical chloride permeability, so that trans-epithelial chloride transport is limited by transport across the basolateral membrane of tracheal epithelial cells, which mainly reflects the activity of cotransporters. Nystatin (400 units/mL) increased the base level of trans-epithelial 36CL flux by approximately 1.5 times and eliminated the UTP stimulation of that flux. Nystatin treatment also eliminated saturable basolateral [3H] bumetanil binding UTP stimulation, a measure of functional Na-K-CL cotransporters in these cells; The stimulation of binding by isoprene was only slightly inhibited by nystatin treatment [2]. Nystatin significantly enhances endostatin uptake by endothelial cells through endostatin internalization, which is mainly converted to clathrin mediated pathway. Nystin-enhanced endostatin internalization also increases its inhibitory effect on endothelial tube formation and migration [3].
In vivo studies, the combination of nystatin and endostatin selectively enhanced endostatin uptake and biological distribution in tumor blood vessels and tumor tissues, but failed to enhance endostatin uptake in normal tissues of tumor-bearing mice, ultimately leading to increased antiangiogenesis and anti-tumor efficacy of endostatin in vivo [3]. Lipogenic nystatin, at doses as low as 2 mg/kg bw/day, protected neutropenia mice in a statistically significant manner against Aspergillus induced death at a 50-day time point compared with no treatment, normal saline, or salt-free treatment.
